Differentially expressed genes (DEGs) were analysed through three transcriptomic datasets of GSE29409, GSE59034 and GSE72158 from the GEO database regarding subcutaneous adipose tissue (SAT) after BS, and 37 DEGs were identified. The weighted gene co-expression network analysis (WGCNA), last absolute shrinkage and selection operator (LASSO) logistic regression and support vector machine-recursive feature elimination (SVM-RFE) algorithms further screened four key genes involved in the regulation of STMN2, SFRP4, APOE and MXRA5. The GSE53376 dataset was used to further confirm the differential expression of SFRP4, APOE and MXRA5 in the postoperative period. GSEA analysis reveals activation of immune-related regulatory pathways after surgery. Finally, the silencing of MXRA5 was found by experimental methods to affect the expression of PPARγ and CEBPα during the differentiation of preadipocytes, as well as to affect the formation of lipid droplets. In conclusion, SAT immunoregulation was mobilized after BS, while MXRA5 was involved in the regulation of lipid metabolism.", "claims": [{"id": "claim-method", "statement": "A co-expression network was formed through the WGCNA R package [27] in order to find the most relevant modules to the phenotype.", "kind": "methodological", "target": null, "evidence": [{"asset": "source:pmid:36036283", "locator": "sections[5] \"Screening and confirmation of biomarkers\" paragraph 2", "quote": "A co-expression network was formed through the WGCNA R package [27] in order to find the most relevant modules to the phenotype."}], "state": "complete"}, {"id": "claim-data-access", "statement": "Detailed data on this study can be found in GEO database (http://www.ncbi.nlm.nih.gov/geo), reference numbers [GSE29409, GSE59034, GSE72158, and GSE53376].", "kind": "methodological", "target": null, "evidence": [{"asset": "source:pmid:36036283", "locator": "sections[25] \"Data availability statement\" paragraph 2", "quote": "Detailed data on this study can be found in GEO database (http://www.ncbi.nlm.nih.gov/geo), reference numbers [GSE29409, GSE59034, GSE72158, and GSE53376]."}], "state": "complete"}, {"id": "claim-target-result", "statement": "The weighted gene co-expression network analysis (WGCNA), last absolute shrinkage and selection operator (LASSO) logistic regression and support vector machine-recursive feature elimination (SVM-RFE) algorithms further screened four key genes involved in the regulation of STMN2, SFRP4, APOE and MXRA5.", "kind": "qualitative", "target": null, "evidence": [{"asset": "source:pmid:36036283", "locator": "sections[0] \"Abstract\" paragraph 1", "quote": "The weighted gene co-expression network analysis (WGCNA), last absolute shrinkage and selection operator (LASSO) logistic regression and support vector machine-recursive feature elimination (SVM-RFE) algorithms further screened four key genes involved in the regulation of STMN2, SFRP4, APOE and MXRA5."}], "state": "complete"}], "gaps": []}, "experiment": {"state": "complete", "objective": "The weighted gene co-expression network analysis (WGCNA), last absolute shrinkage and selection operator (LASSO) logistic regression and support vector machine-recursive feature elimination (SVM-RFE) algorithms further screened four key genes involved in the regulation of STMN2, SFRP4, APOE and MXRA5.", "steps": [{"id": "read_published_estimate", "order": 1, "name": "Read the published estimate from the source", "kind": "other", "state": "complete", "blocked_on": null, "description": "The claim, method and evidence span published with the source article.", "duration_hours": null, "parameters": {}, "inputs": ["source:pmid:36036283"], "outputs": ["the published estimate and its evidence"], "instruments": [], "reagents": []}, {"id": "obtain_source_data", "order": 2, "name": "Obtain the source data from the named repository", "kind": "data-preparation", "state": "complete", "blocked_on": null, "description": "The GEO series accession resolves and its sample accessions are listed.
